Phylum Bacillota (Firmicutes), Class Bacilli, Order Caryophanales, Family Bacillaceae, Genus Bacillus, Bacillus dafuensis Zheng et
al. 2023.
Species description is based on a single isolate.
Gram-positive, straight rods, 0.9-1.2 x 2.4-4.8 µm in size. Motile. Ellipsoidal
endospores are observed at terminal position.
After growth at 30 ºC for 48 h on Nutrient Agar, colonies are circular, pale yellow, moist,
smooth surface, and about 2 mm in diameter. Growth occurs at 15-50 ºC (optimum 35
ºC), pH 6.0-10.0 (optimum 8.0), and 0-4.0% (w/v) NaCl (optimum 0%). Aerobic.
Isolated from a forest soil sample in Dafu County, Chengdu City, Sichuan Province, China.

Positive results for acid and alkaline phosphatase, catalase, aesculin hydrolysis, esterase (C4), esterase lipase (C8), leucine
arylamidase, naphthol-AS-BI-phosphate hydrolase, alpha-glucuronidase, N-acetyl-glucosamine and Voges-Proskauer test.

Negative results for chymotrypsin, citrate utilization, gelatin hydrolysis, beta-galactosidase, nitrate reduction, starch hydrolysis,
oxidase, valine arylamidase, acid production from: glycerol, D-galactose, D-glucose, D-ribose, D-mannitol, D-sorbitol, xylitol and the
other API 50 CH substrates.
