Phylum Pseudomonadota (Proteobacteria), Class Gammaproteobacteria, Order Pseudomonadales, Family Pseudomonadaceae,
Genus Azomonas, Azomonas macrocytogenes (Jensen 1955) New and Tchan 1982.
Basonym: Azotobacter macrocytogenes Jensen 1955.
Gram-negative or Gram-variable, large, ellipsoidal- to rod-shaped cells depending on
age, 2.0 x 2.5–3.5 µm, occuring singly, in pairs, or in clumps. Distortion of cells, with
irregular, filamentous forms predominating may occur. Motile by polar flagella. Cysts
are not formed but saclike capsular structures may occur and be mistakenly identified
as germinating cysts.
Colonies are 1-6 mm in diameter after 1 week of incubation; initially, they may be
coarsely wrinkled but later they become smooth. On sucrose agar produces large
and moist colonies due to the formation of homopolysaccharides. Produce a
yellow-green visible, blue-white fluorescent pigment (azoverdin) when grown in
iron-deficient medium. Azoverdin is produced in Fe-limited cultures grown at 28 ºC but
not at 34 ºC. Optimum temperature for growth is approximately 30 ºC; minimum 14 ºC.
Variable growth at 9 ºC. Grows aerobically and microaerobically.
Isolated from soil. Nitrogen-fixing organism.

Positive results for catalase and oxidase.
Can utilize: fructose, glucose, sucrose, acetate, pyruvate, fumarate, malate, succinate, alpha-oxoglutarate, lactate, DL-gluconate,
acetyl methyl carbinol, D-galactose, melezitose, mannitol, maltose and sorbitol.
Negative results for starch hydrolysis and urease.
No utilization of: raffinose, glutarate, benzoate, caproate, malonate, and mucate.
Variable utilization of trehalose.
